Runers is a top-down roguelike dungeon shooter in which the player explores a labyrinth, fights monsters and bosses, and collects runes. Death ends a run, while the rooms, enemies, events, and bosses on each floor are procedurally generated, so each attempt plays out differently.

Runes can be combined into spells and alter their properties; there are 285 spells to discover and upgrade. The player chooses a race and class, then selects one of several random traits when leveling up to strengthen the character. Discovering spells adds entries to the Runedex, and defeating enemies fills out the Bestiary. Five difficulty levels are available.
